PRILOS acts by reducing the gastric acid secretion through its selective inhibition in the enzymatic system of H+, K+ – ATPase
(the proton pump in gastric mucosa) on the surface of gastric parietal cell.

DOSAGE AND ADMINISTRATION
It is recommended to be taken before breakfast.
For duodenal ulcer, gastric ulcer and erosive/ulcerative reflux oesophagitis : 20 mg once daily, should be taken with
In patients with duodenal ulcer, healing should occur within 2 weeks. For those patients who may not be fully healed
after the initial course, healing usually occurs during a further 2-week treatment period.
In patients with gastric ulcer or erosive/ulcerative reflux oesophagitis, the treatment and recovery may take 4 weeks.
Patients who may not be fully healed, may require a further 4-week treatment period.
In poorly responsive patients, 40 mg once daily is recommended and healing is usually achieved within 4 weeks in
patients with duodenal ulcer and within 8 weeks in patients with gastric ulcer and erosive/ulcerative reflux oesophagitis.
Zollinger-Ellison Syndrome : initial dose : 60 mg once daily
The dose should be adjusted to each individual and treatment should be given as long as clinically indicated. Patients
with severe disease may require 20 – 20 mg of omeprazole daily. Dosages more than 80 mg should be given twice
Patients with liver and renal disorder : no dose adjustment required.
Children : clinical experience in children have not been established.
Elderly : no dose adjustment required.
Rare : nausea, headache, diarrhea, constipation, abdominal pain and skin disorder.
WARNING AND PRECAUTIONS
For the treatment in gastric ulcer, the possibility of malignancy should be excluded as treatment may alleviate
symptoms and delay diagnosis.
Should not be given to pregnant and breastfeeding women unless clearly needed.
Patients with hypersensitivity to omeprazole and other components of this drug.
PRILOS may prolong the elimination of diazepam, warfarin and phenytoin, drugs that are metabolized by oxidation in
the liver. Monitoring of patients receiving these drugs is recommended to adjust the dose.
No interactions with propanolol or theophylline but there may be interactions with other drugs also metabolized via the
cytochrome P-450 enzyme system.
No interactions with concomitantly administered antacids have been found.
